Product Description
Custom DNA Primers from Invitrogen are synthetic oligonucleotides made to order with your specified sequence for use in a variety of applications from PCR and sequencing to probes for gene detection. Custom DNA Primers are available as standard deoxynucleotides, modified bases, 5´ modified nucleotides (including fluorescent dyes, enzyme conjugates), and S-oligos for antisense studies. Five scales and four purity levels are available. Invitrogen uses our unique and proprietary technologies and an optimized production process to deliver quality Custom Primers, quickly and efficiently.

The process includes easy ordering, linkage to integrated workstations for guaranteed accuracy, high-throughput synthesis, monitoring by a series of stringent quality control steps, useful product packaging, and overnight shipping for rapid, reliable delivery. Custom oligonucleotides are synthesized on automated, proprietary DNA synthesizers using standard cyanoethyl phosphoramidite chemistry. Consistent oligonucleotide quality is ensured by in-process trityl monitoring and quality control analysis. A comprehensive Certificate of Analysis accompanies each order, indicating quantity, molecular weight, extinction coefficient, sequence, and melting temperature.

Additional Features of our Unique Process

  • Flexibility in ordering: E-mail, Internet, or Fax.
  • Primers are available in two formats: lyophilized in 2-ml microcentrifuge tubes or lyophilized in 96-well arrays for automated operations. A minimum of 48 primers must be ordered for the 96-well array format. Please call us if you require large orders of Custom Primers, or alternate formats such as 384-well plates.
  • Final testing of statistically selected primers by capillary electrophoresis or by mass spectroscopy to further ensure the quality.
  • Certificate of Analysis to include primer name; sequence and length; average coupling efficiency; melting temperature; GC content; amount in ODs, µg, and nmole; and molecular weight and extinction coefficient calculated from your exact sequence, including modification.
  • Label information on each vial is durably printed on clear transparent stock to allow full view of contents.
  • Fluorescent primers shipped in amber tubes to protect light sensitive primers.
  • Technical Services technical support for assistance with primer design, applications, and experiment troubleshooting.
  • Shipping in specially designed, easy-to-identify envelopes by express overnight delivery.
  • A primer reassay service is available free of charge for diagnosis of any primer by OD analysis, mass spectroscopy, or capillary electrophoresis. (Contact Technical Services.)
Scale of Synthesis and Application Purity Guide Starting Scale vs. Final Yield When ordering custom primers, remember that the scale of synthesis (10 nmole, 50 nmole, 200 nmole, 1 µmole, or 10 µmole) is the starting point for synthesis, not the guaranteed final amount. Oligonucleotide length, sequence, GC content, and coupling efficiencies can vary the yield of nmole amounts post-synthesis. Invitrogen guarantees the total yield of oligonucleotide as a minimum number of OD units. Purification by HPLC, PAGE, and Cartridge will also affect final product yield.
