RNA interference (RNAi) was initially proven to be a powerful method of knocking down gene activity by the introduction of a long double-stranded RNA (dsRNA) molecule in invertebrates such as Drosophila and C. elegans. However, in most mammalian cells the introduction of long dsRNA initiates a cellular interferon response that ultimately causes cell shutdown and leads to apoptosis. In mammalian cells the introduction of small 21-23 nt sequence-specific RNA duplexes can act to initiate post-transcriptional gene knockdown and avoid triggering this non-specific effect in mammalian cells.
The siRNA is then transfected into cells. The siRNA associates with an intracellular multi-protein RNA induced silencing complex (RISC). This complex recognizes and cleaves complementary cellular mRNA. The cleaved mRNA is targeted for degradation, ultimately leading to knock down of post-transcriptional gene expression in the cell.
