Protein A covalently coupled to agarose has been extensively used as an affinity purification support for the isolation of a wide variety of IgG molecules from several species of mammals. A typical elution profile of mouse IgG on Immobilized Protein A is shown in the figure below. Pierce offers Immobilized Protein A in several different package sizes. This is highly-purified Protein A, covalently coupled to cross-linked beaded agarose. Protein A is also provided immobilized on UltraLink Biosupport Medium or Trisacryl GF-2000. The latter two supports provide a stable affinity support that will withstand the higher throughput volumes experienced during large scale chromatographies. In addition, because both are hydrophilic matrices, there is little concern with nonspecific binding. For the ultimate in convenience, Pierce recommends its ImmunoPure (A) IgG Purification Kit. This kit contains everything you need to isolate IgG from mouse ascites fluid (or from the serum of various other species). The exclusive ImmunoPure Buffer System enhances the binding of IgG to Immobilized Protein A. The columns in the kit can be regenerated at least 10 times without a significant loss of binding capacity.
