  • Description: Products and Services Bioremediation is a process that uses microorganisms, bacteria, fungi, green plants, and their enzymes to restore the environment to its original state after contamination. Bioremediation has been used for centuries after corruption of soil by salt, sewage, and other contaminants, particularly in agriculture. Bioremediation of soil or water following contamination by crude oil is performed by a variety of bacteria that feed on hydrocarbons, breaking down the petroleum molecules into less harmful substances. These naturally occurring bacteria can potentially be isolated and expanded to help remediate oil spills, an approach well accepted by nature with minimal threat to marine ecosystems.

    Altogen Labs successfully isolated oil-degrading microbes from polluted soil near Galveston, Texas. These microbes were shown in laboratory tests to be highly effective in bioremediation of petroleum-saturated earth. A method was developed to cultivate oil-degrading bacteria specific to the site of a particular spill, microbes that act on the hydrocarbon molecules present at the polluted site. The bacteria are grown in large volumes in an aqueous solution, from which the water is eventually evaporated to yield a high concentration of bacteria in dry form and low volume.

    The product is very stable and the dry bacteria can be activated by simply re-introducing a water-based solution. The growth conditions for these natural bacterial strains were optimized for effective oil degradation in Texas soil. Optimization included considering variables in temperature, soil conditions, and oil composition such that these microbes could be used to remediate contaminated soil on-site when reintroduced in large numbers. This technology offers great potential for cleaning up contaminated soil due to rupture of underground storage tanks, pesticides, percolation of contaminated surface water, oil and fuel dumping, leaching of wastes from landfills, and direct discharge of industrial wastes to the soil.

    However, despite its toxicity to most organisms, a considerable percentage of petroleum entering marine systems is eliminated by the hydrocarbon-degrading activities of various microbial communities, in particular, by a recently discovered group of microbes known as hydrocarbonoclastic bacteria (HCB). Of this group, the bacterium Alcanivorax borkumensis was the first HCB to have its genome sequenced by scientists and is now the subject of study by researchers around the world.

In addition to standard ribonucleotides, company scientists also incorporate modified nucleotides (e.g., aminoallyl-UTP) to produce labeled RNA probes and amplifies the original mRNA more than 1 million-fold. RNA is also purified and prepared for all additional applications.

    Gene Knockdown by shRNAs
    The customer provides the name and accession numbers of the target genes, mails the cells and primary antibodies for Western blotting analysis to Altogen Labs, and the lab team does the rest .

    Altogen’s knockdown service includes design, construction, and confirmation of the 3'-5' sequence the shRNA vectors for every target gene. Design and synthesis of the SYBR-labeled qPCR primers for quantitation of target gene expression, shRNA transfection into the cell line supplied by the customer, and targeted gene expression analysis by qPCR at 24, 48, and 72 hr post-transfection and Western blot at 48 and 72 hr after transfection are performed by company scientists.

    Altogen Labs guarantees 70% or more inhibition of target gene expression. Transfection controls includes the negative vector control with scrambled sequences (21 bp) and a GFP protein vector control for assessment of transfection efficiency.
