Custom siRNAs can be synthesized according to the information you supply or can be custom designed by the company’s experts. Numerous options are possible, including various modification combinations, synthesis scales, and purification levels. Labels and modifications include amine, biotin, fluorescein, phosphorylation, and Cy-dyes, in lengths ranging from 19 to 45 mers, in single-stranded or duplex formats. Regarding QC, most companies use MALDI-TOF and PAGE, with typical purity levels of 80–90%. HPLC purification is often available (usually for an extra charge) for experiments requiring higher purity, and additional steps are often in place to ensure proper strand annealing. Newer synthesis technologies that give higher coupling efficiency and faster deprotection are of particular interest.
