Description
Klenow Fragment is a sequencing grade 68KDa proteolytic subfragement of E. coli DNA polymerase I, obtained by subtilizing cleavage of the haloenzyme. It lacks exonuclease activity and produces blunt ends. The fragment can be used to synthesize double-stranded DNA from single-stranded templates, second strand cDNA fragments, filling in 5′ ends and recessed 3′ ends of DNA fragments and radio labeling and dideoxy sequencing.
