Melon Gel uses a mild buffer system that enables functional antibody isolation from ascites, cell culture or serum samples using a simple flow-through procedure. Melon Gel binds non-relevant proteins, allowing the purified antibody to be isolated at physiological conditions suitable for storage and downstream applications. This flow-through purification is especially useful for antibodies that lose their antigen-binding activity at low pH. This robust procedure offers higher recovery and purity than bind-and-release methods. Typically, lgG can be isolated within 15 minutes using Melon Gel Spin Plates. Each well of the 96-well plate contains 100 µl of Melon Gel, which can process 10-100 µl of samples and purify up to 1 mg of IgG per well. The Melon Gel approach to antibody screening and purification requires cell culture supernatant, ascites and serum samples to be desalted or diluted in the purification buffer before processing. Zeba 96-Well Desalting Spin Plates are useful for this application.
