Description

After you have identified your target from a microarray, validated results using qPCR or RNAi, confirm the proper expression or accumulation of each component by western blot analysis and/or immunohistochemical (IHC) staining. Validated BioModule™ units are available for both of these applications: Western blots allow you to detect expression of protein(s) in a complex mixture of biomolecules using probes such as an antibody or streptavidin specific to a protein or a protein tag such as a histidine, biotin, c-myc, or FLAG. The process involves sample preparation, gel electrophoresis, transfer, blocking, probing, and detection with either colorimetric, chemiluminescent, or fluorescent detection. BioModule™ Western provides the major reagents and protocols necessary for each of the steps for protein assessment   •   Immunohistochemical (IHC) staining allows you to directly detect antigens in tissue fixed on a glass slide. Indirect methods, such as those provided in the BioModule™ IHC, involves an unlabeled primary antibody which reacts with tissue antigen, and a labeled secondary antibody which reacts with the primary antibody. The secondary antibody must be against the IgG of the animal species in which the primary antibody has been raised. This method is more sensitive than direct staining, due to signal amplification through several secondary antibody reactions with different antigenic sites on the primary antibody.™ IHC Staining for tissue offers the core reagents you need for general immunohistochemical applications.
