BioModule™ Microarray Units includes major reagents required to purify your RNA, synthesize and label cDNA, then subsequently hybridize and wash your array. BioModule™ Microarray is available with SuperScript® Plus cDNA Labeling System optimized with Alexa Fluor® dyes to yield proven cDNA labeling results to support direct and indirect labeling methods. All BioModule™ Microarray units have been validated to ensure that they are compatible across your experiment.BioModule™ Microarray unit contains TRIzol® Reagent for RNA isolation and a PureLink™ Micro-to-Midi™ Total RNA Purification Kit for post-extraction clean up to ensure high-quality total RNA. The PureLink™ Kit is a one-hour procedure that requires no handling or disposal of organic waste. RNA binds to a silica membrane and impurities are removed by washing through vacuum filtration or centrifugation. High-purity total RNA is eluted into RNase-free water, ideal for immediate use in labeling.indirect labeling, mRNA or total RNA is reverse transcribed using SuperScript® III RT, incorporating amino-modified dUTP and dATP into the synthesized cDNA. The template RNA is then degraded by base hydrolysis, and the reaction is neutralized with acid. The amino-modified cDNA is then purified to remove unincorporated nucleotides, primers, and buffers. In the second step, the modified cDNA is coupled with the active form of Alexa Fluor® 555 or 647 fluorescent dye. The labeled cDNA is purified with a low-elution volume spin column to remove any unreacted dye. The resulting cDNA is ready for hybridization to microarrays.direct labeling, an anchored oligo(dT) anneals to the mRNA template. SuperScript® III Reverse Transcriptase (RT) extends from the priming site, incorporating dUTP labeled with Alexa Fluor® dye directly to produce labeled cDNA. Template RNA is degraded by base hydrolysis.
