SulfoLink Coupling Gel is an exceptionally useful activated gel support for covalent immobilization of cysteine-containing proteins and peptides. The iodoacetyl-activation ensures efficient and stable reaction to sulfhydryl (–SH) groups present in the side chain of reduced cysteine residues. The SulfoLink Chemistry is especially useful for preparing affinity columns to purify antibodies raised against immunogens that were constructed by conjugation of cysteine-containing peptides to maleimideactivated carrier proteins. By immobilizing the peptide through the same functional group as was used to conjugate it to the carrier protein, one ensures its proper orientation at the end of a long spacer arm for optimal antibody binding. Specific orientation of antibodies and other proteins may also be accomplished using SulfoLink Coupling Gel. For example, hinge region disulfide bonds may be selectively reduced with 2-mercaptoethylamine (included in the SulfoLink Kit) so that functional halfantibodies may be immobilized in a manner that ensures that antigen binding sites remain available and functional.
Two iodoacetyl-activated supports are available. SulfoLink Coupling Gel is 6% cross-linked beaded agarose, suitable for small-scale batch methods and gravity-flow column purification. Kits and stand alone coupling gel are available. UltraLink Iodoacetyl Gel is activated UltraLink Biosupport, a durable support suitable for medium pressure chromatography and FPLC.
