The large fragment of DNA Polymerase I from E. coli, Klenow enzyme retains polymerization and 3→5 exonuclease activity but lacks the 5→3 exonuclease activity of intact DNA Polymerase I. It is well-suited for the conversion of 5-protruding ends to flush ends for
cloning(1), in 3-end labeling of DNA fragments for sequence analysis(2) for use in the DNA sequencing method of Sanger, et al.(3), for second-strand synthesis of cDNA(3) and for random primed labeling of DNA fragments.
