Description
The Blood Dendritic Cell Enumeration Kit was developed for easy identification and counting of dendritic cells and dendritic cell subsets in whole blood or PBMCs by flow cytometry. It is based on the dendritic cell-specific surface antigens CD303 (BDCA-2), CD141 (BDCA-3), and CD1c (BDCA-1).1 The markers allow easy identification of three distinct dendritic cell subsets in blood: plasmacytoid dendritic cells (PDCs) are identified by CD303 (BDCA-2), type-1 myeloid dendritic cells (MDC1s) are identified by CD1c (BDCA-1), and type-2 myeloid dendritic cells (MDC2s) are identified by CD141 (BDCA-3) expression. The distinct dendritic cell subsets are analyzed within one sample by three-color (PDCs and MDC1s) or four-color (PDCs, MDC1s and MDC2s) flow cytometry.
The kit includes all reagents needed for a complete analysis: pre-mixed antibody cocktails for dendritic cell detection and for isotype control, a dead cell discrimination dye, and ready-to-use reagents for erythrocyte lysis and cell fixation. The standardized reagents and protocol ensure reproducible results.2 The dead cell dye used in this kit allows staining of dead cells in combination with cell fixation. This is particularly useful when working with hazardous samples.
The assay requires only small samples (either 300 µL of whole blood or 1×106 PBMCs, both for sample and control).
