Description
The BLOCK-iT™ RNAi TOPO® Transcription Kit is a high-yield RNA synthesis kit that results in pure RNA transcripts. These transcripts are compatible with the BLOCK-iT™ Dicer Kits for mammalian cell RNAi analysis. They can also be used directly to analyze the effects of endogenous gene knockdown in organisms that do not have an interferon response, such as most non-mammalian eukaryotes, including insects (Figure 1), nematode, trypanosomes, and some mammalian stem cell models.
The BLOCK-iT™ RNAi TOPO® Transcription Kit method facilitates fast and easy generation of dsRNA through the addition of T7 TOPO® linkers to PCR products containing a gene of interest, without the need for special primers (Figure 2). The specific mixture of components assures that the high levels of sense and antisense transcripts made in separate reactions will be of the quality needed for efficient and reproducible knockdown of endogenous gene expression.
