Description
ThermoScript™ Reverse Transcriptase (RT) is an avian RT with reduced RNase H activity designed for RT-PCR (1). It is engineered to have high thermal stability (up to 70°C) (Figure 1), copy RNA with secondary structure, and improve priming specificity (2). ThermoScript™ Reverse Transcriptase offers:
- Greater first-strand cDNA yields than reverse transcriptases with RNase H activity
- More full-length cDNA synthesis than reverse transcriptases with RNase H activity
- Flexibility–uses either total or poly(A) + RNA
- The ability to use RNA primed with oligo(dT), random primers, or a gene-specific primer
- Generation of RT-PCR products up to 12 kb
Application: Synthesis of first-strand cDNA for RT-PCR (1).Unit Definition: One unit of ThermoScript™ is the amount of enzyme required to incorporate 1 nmole of deoxyribonucleotide into acid-precipitable material in 10 min. at 37°C using poly(A)•oligo(dT)25 as template•primer.
