I used to measure the concentration of Hsp71 in my human PBMCs to check for their values before and after treatment with some X (X because its still a secret pro-drug) drugs which serve as antagonists for PRR receptors. We do see changes and that's why we used this kit to check for the levels of heat shock protein 70. We used also antibodies but were not so happy for them (didn't work very well).

Source was Human PBMC cells treated with drugs
Short protocol (from the company)1. Prepare all reagents, samples and standards;2. Add 100µL standard or sample to each well. Incubate 1 hours at 37°C;3. Aspirate and add 100µL prepared Detection Reagent A. Incubate 1 hour at 37°C;4. Aspirate and wash 3 times;5. Add 100µL prepared Detection Reagent B. Incubate 30 minutes at 37°C;6. Aspirate and wash 5 times;7. Add 90µL Substrate Solution. Incubate 10-20 minutes at 37°C;8. Add 50µL Stop Solution. Read at 450nm immediately.
Substrate solution is sensitive on light!
I got lower Hsp70 levels on the treated cells with the drug, and thus good data for publishing and show the effects of the X drug.
