C57BL/6 mouse lymph node and splenic cells

Protocol Overview

Spleens and lymph nodes from C57BL/6 mice were collected, turned into a single cells suspension, and then run through the R&D Systems Mouse CD4+ T Cell Enrichment Column in order to generate a 90% plus CD$+ T cell population. This population was then cultured in vitro for 5 days with a milieu of cytokines and antibodies in order to generate either a Th1 or Th17 skewed phenotype.

Tips

Do not mix the lymph node and splenic cells together until after the column as it will cause clumping of your cells.

Results Summary

The column generates 90% or more pure population of CD4+ T cells every time it is used.
