This stain can be detected on a red laser with a 780/60 bandpass filter. It fits well in complex flow cytometry staining panels with only modest compensation requirements. It maintains a bright and distinct stain in various fixation procedures. Used at a dilution this live/dead marker can be well worth the cost.

Protocol Overview

Stain up to 3x10^6 cells in 50ul of PBS with 1% FBS. Use the Fixable Viability dye eFluor 780 in an antibody cocktail at a 1:1000 dilution for 30 minutes at 4 degrees Celcius. Wash samples and proceed with desired fixation and any subsequent staining steps.

Tips

It may be able to titrate out more than 1:1000 but this gives a clear distinct population.

Results Summary

It will maintain a robust stain despite fixation or numerous washes. Dead cells are easily visible using this dye.
