The purpose is to isolate pure CD8 T cells (untouched) for adoptive transfer into mice for an in vivo study.

1. Total splenocytes were prepared by grinding with nylon mesh (70 um) 2. RBCs were removed by treating with ACK buffer 3. Cells were further isolated for CD8 cells according to manufacturer's protocol
During the procedure, it is important to remove any debris or aggregates.
The quality was comparable to kits from other companies such as Miltenyi Biotec, StemCell Technologies and BD Biosciences.
