The purpose of using this kit is to purify the "Untouched" CD4 T cells from murine spleen and lymph nodes. Isolated T cells were cultured in vitro or were transferred into mice for in vivo study.

Protocol Overview

1. Total splenocytes and lymph node cells were prepared by grinding with 70um nylon mesh. 2. RBCs were removed by ACK lysis3. Add Ab cocktail and incubate for 15-20 min at 4 degrees 4. Add magnetic beads and incubate for 15-20 min at 4 degrees 5. Put the tube in the magnetic stand for 5 mins 6. Non-attached cells (CD4+) were taken
