We wanted to determine whether there were differences in TNF-a protein in mouse brains in 2 different mouses models after treatment with a chemical that induces demyelination.

The kit contained all of the necessary components for a standard sandwich ELISA. We coated a 96 well plate with the capture antibody overnight and used the recommended reagent diluent for all dilutions (1% BSA in PBS). We added the sample and stds and allowed to incubate overnight. The next day we added the detection antibody for 2 hours, added strep-hrp, followed by TMB solution for 10 mins. After the 1N sulfuric acids was added to stop the reaction the plate was read at 450nm.
Incubate samples overnight is low amounts of TNF-s is expected
Reproducible results. We were able to detect TNF-a in all our samples
