CellTiter Glo is a great assay to determine cytotoxicity of drugs/agents. Reagents are resistant to many compounds that may interfere with such reagents of other cell viability assays.

Human cancer cell lines
Cultured cells in 96-well plates were treated with compound(s) for certain amount of time before addition of reagent. Incubated for 10 minutes and read plate on luminometer.
Make sure cells are properly lysed and ensure proper timing between reagent addition and reading on luminometer
Cytotoxicity of compound is determined by comparing luminescence counts of treated cells with those of untreated control cells.
