The goal was to elucidate the molecular mechanism of Pkm2 by overexpressing it in aggressive cancers. To rule out the mechanism, we used core molecular biology techniques like by generating CRISPR knock out cell lines. We then have to validate the cell line by doing immunoblot analysis. Generally, PkM1 protein is expressed constructively and executes its enzymatic function, whereas Pkm2 is reported to be induced at cancer disease condition only.
