Our lab studies HIV infection in transgenic and humanized mouse models. To detect HIV infection we isolate RNA from different mouse tissues and do real time PCR to quantify copies of viral RNA. Because RNA can be very unstable and degrade quickly at room temperature we use this reagent to stabilize the RNA while we extract it from tissue.
RNA extraction
Mouse tissue
Determine appropriate amount of reagent for tissue of interest. Collect the tissue and submerge it in reagent. Store the sample per manufacturer's instructions. Proceed with RNA extraction using the Qiagen kit (we have only tested this reagent in combination with qiagen RNA extraction kit).
Cut the tissue into slices less than 0.5cm thick before submerging
Good yield and quality of RNA extracted. Better than when no RNA stabilization reagent used.
