Lipofectamine 3000 can be used to transfect a broad spectrum of cell line with nucleic acids. Transfection of cells with plasmid DNA is well optimized, and the procedure is simple and quick. Depending on the cell line, there may be some cytotoxicity associated with the reagents.

Protocol Overview

Lipofectamine 3000 comprises of two reagents: Lipofectamine 3000 and P3000. The first reagent is placed in a tube with OPTIMEM media. A second tube contains P3000 with OPTIMEM, and DNA of choice. Contents of both tubes are combined and mixed prior to incubation for 15 minutes. This mixture is added to cells to allow for tranfection to occur.

Tips

If transfecting many wells, make sure to have extras to account for pipetting error.

In general, I got efficient transfection of cells with DNA in my cell lines.

Lipofectamine 3000 is a great tool for transfection of plasmid DNA with little concern for cytotoxicity in many cell lines. Some cell lines may be too sensitive to this method, so another tool must be used.
