Company:

Invitrogen Antibodies

Product Name:

CD185 (CXCR5) Monoclonal Antibody (SPRCL5), Biotin, eBioscience™

Catalog Number:

13-7185-82

We work to identify the T follicular helper population in an autoimmune setting using this antibody for CXCR5 staining. Streptavidin conjugates provide the bright color we needed to identify the population while direct fluorophore conjugates didn't work as well for our experiments.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Murine Lymphocytes

Primary Incubation

1:50 dilution, 1 hour at room temperature incubation

Blocking Agent

FC Block

Secondary Incubation

1:100 Streptavidin dilution, 30 minutes at 4 degrees in the dark incubation

Tertiary Incubation

None

Detection

BD LSR II

Results Summary

CXCR5 on T cells appears as a branching population from the negative population. A fluorescence minus one can be used to detect the negative population.

Summary

The Good

It gives the bright signal needed to identify CXCR5.

The Bad

It can be challenging to optimize.

The Bottom Line

You can identify CXCR5 more efficiently with this two-step staining method then with direct fluorophore conjugates.
