We are studying a newly discovered receptor called transcobalamin receptor (TCblR) encoded by CD320 gene. This receptor is required for uptake of cobalamin in humans as well as mouse. To determine where it is expressed and how it is involved in materno-fetal transport of cobalamin, we studied its expression by immunohistochmeistry using this secondary Ab.

Experimental Design and Results Summary

Applications

Immunohistochemistry

Sample

Paraffin embedded mouse placenta sections

Primary Incubation

1:200, 1 hour at room temperature

Blocking Agent

Normal goat serum

Secondary Incubation

1:1000, 1 hr at room temperature

Tertiary Incubation

NA

Detection

DAB staining

Results Summary

We observed expression of TCblR on the fetal side of placenta, known as labyrinth. The expression was highly specific to this region in WT mouse. We used a TCblR KO mouse as a control and found no expression with immunostaining using the same protocol.

Summary

The Good

Works very well even at higher dilutions

The Bad

Incubation has to be at least 1 hr

The Bottom Line

We have used this Ab with many other primary Abs for mouse sections it it always gives wonderful results.
