We are studying a KO mouse model of cobalamin deficiency and do a lot if immunohistochemistry using paraffin embedded brain sections. We used this kit to determine the expression of a presynaptic protein in the hippocampal region.

Paraffin embedded mouse brain sections
Prior to immunostaining, we performed antigen retrieval using citrate buffer consisting of 10mM, pH 6.0 with 0.05% Tween 20. Sections were treated with citrate buffer for 40 minutes at 90–100°C. After 40 minutes, sections were let stand at room temperature for another 40 minutes and were washed with PBS afterwards. We treated the sections with 3% hydrogen peroxide for 5 min and then started the immunostaining protocol as described by the manufacturer. We incubated the sections with the primary Ab for one hour. Counter-staining was done with hematoxylin and sections were dehydrated in increasing percent of alcohol (70-100%), cleared in xyelenes and mounted with permount.
The blocking reagent is already diluted, no need to dilute it further
We obtained staining (representing expression of our protein of interest) in the stratum radiatum region of hippocmapus.
