Our lab routinely performs immunohistochemistry on both paraffin-embedded sections and frozen sections. We have found that some OCT suppliers do not have quality product and lower quality product affects tissue sectioning. This OCT supplier has consistently worked well for us.

Perfuse mouse with 4% paraformaldehyde, collect tissue, let sit in 4% PFA for 48 hours then 25% sucrose for 48hrs. Place tissue in mold, cover with OCT, flash freeze

Be sure to keep the bottle upside down to ensure no bubbles are introduced into the mold
