We do a lot of whole lung mouse and human flow sorts, so we use this dispase as the base for our tissue digests. We isolate a variety of cell populations and it works well for our needs.

Protocol Overview

For our mouse prep and for liberating cells from matrigel, we use this at a concentration of 15units/ml. Mouse digest is about 45 minutes, cell liberation can take 20-40 minutes for us depending on cell type and number. For our human lung digest, we use a slightly lower concentration but a longer time period. We always make this solution in Hanks BSS and filter sterilize it prior to using for anything.

Tips

Make sure to filter sterilize it prior to use and make it up in Hanks buffer solution!

Results Summary

We've used this dispase reliably for several years now and have solid results. Our flow protocols have been well refined through time and we still prefer this dispase. We've played around with the time, temperature, and basis for our digests, and this seems to be the best enzyme for our needs.

Summary

The Good

Reliable, breaks down lung tissue very efficiently.

The Bad

Sometimes with fibrous tissue you have to vortex it throughout digest to get it going.

The Bottom Line

Works great for our purposes, allows us to have a faster digest that is still gentle on the cells.
