We embed a lot of mouse and human tissue, namely lungs. We do a lot of immunofluorescence and choose to use frozen tissue because of convenience and practice with it. This particular brand of OCT is the only one that we like-we've tried others and the way the blocks cut and form just doesn't compare.

Protocol Overview

We run a full prefixation protocol (1 hour PFA, subsequent PBS washes, sucrose incubation) and inflate our lungs with a mixture of OCT and PFA for fixing and shape retention. Our final step is cutting the lobes apart and arranging them in a cryomold filled with this OCT. They are then frozen and cut/stained at a later date.

So much immunofluorescence data has been obtained using this OCT-it's difficult to categorize. Every block we make and stain is made and stained with this OCT.
