The goal of this experiment was to expand CMV-specific T cells from human donors. This product was selected because it covered different epitopes for different CMV antigens.
In vitro T cell:DC co-culture
Human monocyte-derived DCs and autologous T cells.
Human moDCs were treated with CMV Antigens at 2 ug/mL overnight. T cells (labeled with proliferation dye) were added the next day and cultured for 4 days. After 4 days, T cells were harvested and assayed for proliferation dye dilution.
CMV antigen dose can be titrated down.
CMV-specific T cells were found to have undergone one round of cell division after the 4 day culture period.
