We wanted to determine whether unactivated bone marrow derived macrophages from our conditional knockout mouse secreted the same amount of GM-CSF as WT mice under the same culture conditions.

Protocol Overview

The kit contains all of the components needed for a standard sandwich ELISA. A capture antibody (anti-Mouse GM-CSF monoclonal antibody), a detection antibody (Biotinylated Anti-Mouse GM-CSF monoclonal antibody), Enzyme Reagent (Streptavidin-horseradish peroxidase conjugate) and standards (Recombinant mouse GM-CSF). The capture Ab was added to each well of a 96 well plate and left overnight at 4°C. Wells were washed 3x before blocking with Assay Diluent for 1 hour at room temperature. The standard or supernatant of my cell culture was added to the wells for 2 hours at room temperature. The working Detector was added to each well followed by the substrate solution and stop solution. The plate was read at 450 nm. The plate was washed 3x with Assay diluent before the addition of each solution.
