Our lab does some proteomic work on proteins expressed in an archaeal model organism. In preparation for certain experiments, such as for LC-MS, we would need to quantify the total amount of protein within the sample. For these reasons we have used the BCA protein assay kit.

Archaeal Crude protein extracts, partially purified extracts (no detergents)

Protocol Overview

Followed provided directions:1. Prepare BSA standards2. Combine provided reagents and add to protein samples3. Incubate for 30 min at 30˚C. 4. Measure absorbance at 562 nm.5. Determine protein concentrations based on BSA standard curve.

Tips

Color will start to change immediately once working reagent is added, even at room temp. If measuring many samples (eg. on microplate), add all protein samples first, then use multichannel to add working reagent all at once.
