We were supplied with Lot No: B215037. Upon doing a QC check of the protein by ELISA using the anti-His antibody (R&D) and anti-mouse HRP (both at 1:1000 dilution), we didn’t get any signal. Looks like the His-tag (present at the C-terminus) is lost, thus protein is clearly not what the company claims it to be. Upon contacting the distributor for a refund or replacement, we got the following reply: "The product is guaranteed to work as listed on the datasheet specifically.  In this case, it’s guaranteed active to be used in a bioassay, but does not list WB/ELISA so it may not work in those applications."

Protocol Overview

Wells coated at 5ug/ml with Protein-His or MMP2-His at 4C, overnight. Primary Ab added: anti-His (R&D, MAB050R) at 1:2000, incubated at room temperature for 60 mins Secondary Ab added: anti-mouse IgG1 (Cell signalling, 7076S) at 1:2000; incubated at room temperature for 30 mins Developed with substrate:peroxidase mix at 1:1 Reaction stopped with 1M Phosphoric acid
