This antibody (2186) was used to perform immunofluorescence microscopy on HeLa cells to detect endogenous CENP-A, also known as the chromatin-associated protein CSE4 (capping-enzyme suppressor 4-p). 2186 showed moderate immunoreactivity that was consistent with the centromere. Background levels were also moderate. This antibody is a good choice for multicolor microscopy.
Immunofluorescence
HeLa cells fixed in 3.7% paraformadehyde
1 hour at room temperature at 1:100 in blocking agent
PBS in 1% normal goat serum and 0.1% triton X-100
Goat anti-rabbit conjugated to Alexa 568

Confocal microscopy
This antibody produced moderately strong staining on Hela cells, there was also a moderate level of background staining that was non-nuclear. Overall this antibody appears to be specific to the centromeres.
