Our lab commonly isolates CD4+ T cells from mice to perform in vitro experiments. To isolate this cell population with high purity and yield, Dynabeads are used.
Cell isolation/positive selection of cells
Mouse lymph nodes and spleen
Isolate lymph nodes and spleen from mouse, grind and spin, wash pellet, resuspend pellet in 5ml cell culture media, add 80uL cells, incubate at 4*C with rotation for 35 minutes, pellet beads+cells, wash, resuspend and detach cells from beads
Adjust volume of beads and DETACHaBEADS from the recommended volume suggested by the supplier to maximize cell isolations
A high purity of cells is achieved with large yields and great viability.
