I tried to work out the condition to detect Lipid droplets or lipid-expressing cells within tissue (Frozen cyrosection slide). Life Technologies' LipidTox Green Neutral Lipid staining kit is very easy to follow and gave me the clear lipid staining results.
Lipid staining
20-month-old mouse thymus cyrosection slides (10um)
1. Air dry the slides 1- 5mins.2. PBS ~10-12mins to remove OCT3. Blocking at RT for 1h4. Keratin8 (1:20 red) staining at RT for 1.5h5. 3X PBSwash at RT for 6~8 mins6. 2nd antibody (1:1000) and DAPI (1:1000) at RT for 1 h7. 3X PBS wash at RT for 6~8 mins8. Lipid green (1:200) at RT for 1h9. Mount with Prolong gold antifade.
Protect from light during handing
Detection of clear lipid-expressing cells within thymus and peri-thymus lipid droplets (showing in green in IF picture). Keratin8 (Red)
