The aim of this experiment was to embed sorted thyroid progenitor cells in the basement membrane for differentiation and maturation into thyrocytes. The matrix was recommended by colleagues and is widely used in the literature.

Protocol Overview

Aliquots of matrigel were thawed on ice. Cells were pelleted and then mixed with 20 ul of media. Matrigel was added and mixed with the cells. Matrigel mixture was pipette as droplets in 12 well plate. The plate was incubated for 15-20 minutes and then media was added to the solidified drop.

Tips

Aliquot the matrigel for use to avoid repeated thawing which may introduce bubbles in the gel.

Results Summary

Thyrocytes were well formed and matured in the matrigel matrix.
