Our lab uses HIV molecular clones that express Renilla luciferase reporter. We infect cells in vitro, or perform in vivo experiments where we infect our mice with these viruses. As a read out, we test cell lystaes for their luciferase activity by using this kit.

HIV infected cells
Follow the kit instructions. Cells are lysed in 1X lysis buffer (5X stock in the kit), vortexed well, frozen in -80 degrees for at least 1 hour, then thawed and read in presence of substrate.
Make sure you vortex cells well in you have an infection assays like ours. Otherwise all intracellular virus does not get released in the sup.
Kit works very well. Very clean and sensitive assay.
