QIAGEN's QuantiTect Probe PCR Kit is a reliable qPCR system for our parasite DNA 'TaqMan' assay. Like all QIAGEN kits, it comes with all of the reagents required for a qPCR set-up (excluding template and primer-probe sets). We use the kit to amplify parasite DNA from plasma samples in the absence of gDNA extraction, and this product provides us with consistent, specific results.

Protocol Overview

Prepare a master mix (Probe PCR mix + primers + probe + water) according to the manufacturer's guidelines and add to each well. Add samples (preferably in duplicate or triplicate), including experimental samples and controls, to the plate. Run each plate immediately according to the manufacturer's thermal cycling guidelines -- which are nicely outlined in the manual included in each kit.

Tips

Always run the appropriate controls (NTC, IC, positive controls -- standard curve).

Results Summary

Like all qPCR experiments, raw values are available immediately following each run. We include a standard curve of 5-fold dilutions of parasite DNA (30,000 to 1 genome/ul) to quantify parasite DNA in our experimental samples.

Summary

The Good

Time efficient, simple protocol, compatible with mutiple qPCR systems, reliable/consistent/clean results.

The Bad

Slightly expensive -- similar to all TaqMan/dual-labeled probe systems.

The Bottom Line

I highly recommend this kit for all TaqMan-like qPCR assays that are run on Applied Biosystems qPCR machines. We have had success with this kit in detecting our target from small volumes of plasma (in the absence of gDNA extraction), so I imagine that the kit works incredibly well on isolated/purified gDNA/cDNA as well.
