The present experiment was conducted to test the effect of feeding different types of milk to diabetic person on resting serum T4 concentration. The individuals were allowed to feed a standard diet ad libitum supplemented with a fixed amount of milk samples for six weeks. After end of the experiments, blood was collected, serum was separated and performed ELISA using Abcam kit.

At the end of experiment, blood was collected in tube from individuals, and plasma was separated. Serum was diluted with sterile PBS (1:400) and along with HRP-conjugated Thyroxine serum was added to ELISA plate well. After 60 minutes incubation of serum in ELISA plate at room temperature, wells were washed five times with washing buffer to remove any unbound T4 and HRP-conjugated thyroxine. After that TMB reagent was added and incubated for 20 minutes to allow colour development. The developed colour reaction was stopped was read in a ELISA reader against blank.
