This is a luminescent assay that measures caspase-3 and -7 activities in purified enzyme preparations or cultures of adherent or suspension cells. The assay provides a proluminescent caspase-3/7 substrate, which contains the tetrapeptide sequence DEVD. This substrate is cleaved to release aminoluciferin, a substrate of luciferase used in the production of light. We were looking for a higher throughput way to look at cell death measures in 96-well plates and this was perfect for the job. Increased luminescence is equal to increased caspase activation.

Add Caspase Glo Reagent, mix, and incubate at least 30minutes. Measure luminescense
Use a white walled plate if you can. It gives you a greater and more sensitive signal
We were looking at stressors and treatments to rescue these stressors. We recorded our luminescence and then subtracted the background values from our negative no cell control wells from all other samples. Then we compared our stressors and treatments to our no treatment control to pick out stressors that initiated caspase activation and cell death and the treatments that could prevent this response. Increased luminescence is increased caspase activation
