We have used this antibody successfully on Western blots to look at active caspase 3, this is the one we find to work best. When we were looking for a cleaved caspase 3 marker for staining we gave this one a shot, and although it does work the staining is very weak. We tried to increase the concentration of antibody, but at 1:300 the background began overtaking the weak signal.
Immunofluorescence
Primary Mouse Neurons
1:300 in 5% Donkey Serum at 4C overnight
10% Donkey Serum at RT 1hr
1:400 Dylight Donkey anti-rabbit

Microscopy
The antibody did work, however the staining is very weak even in our positive control. We tried to increase the concentration of antibody, but at 1:300 the background began overtaking the weak signal. If you don't need a lot of sensitivity and are just scoring +/- this marker would be good since itworks so well via Western Blot. If you are tracking intensity changes or more in depth analysis, I would test another marker.
