HL-60 cells are able to differentiate to neutrophils after all trans retinoic acid (RA) treatment. They are also able to differentiate to monocytes after 1,25-dihydroxyvitamin D3 (D3) treatment. A characteristic of this conversion is CD11b expression. Cell culture volumes containing one million cells were spun down and resuspended in 200µl PBS with 2.5µl CD11b Tonbo 20-0118 antibody. This antibody has high background but is suitable for high expression samples.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

HL-60 cells ; untreated, all trans retinoic acid (RA), 1,25-dihydroxyvitamin D3 (D3) treated

Primary Incubation

1 million cells were incubated 1h , 37C, in 200µl PBS with 2.5µl CD11b antibody

Blocking Agent

None

Secondary Incubation

None

Tertiary Incubation

None

Detection

Instrument: BD LSR II. APC was excited at 633 nm and emission was collected through 660/20 band-pass filter.

Results Summary

APC Anti-Human CD11b (ICRF44) Cat.No. 20-0118 Lot c0118081216202 has high background. This makes small changes in expression hard to detect (in our case RA vs C). Samples with high expression (D3 in our case) are still acceptably labeled. We used 2.5ul per test sample compared to 5ul recommended by Tonbo.
