The goal of this experiment was to identify CD25+ T cells in peripheral blood of children with juvenile idiopathic arthritis. Since we only had a limited amount of sample we needed antibodies that worked very well and reliably.

Primary Incubation

50ul of peripheral blood were incubated with the antibody for 30 minutes. Then red cells were lysed using BD lysing solution. After 2 wash steps with 1 ml of PBS, stained cells were resuspended in 300ul FACS buffer and taken directly to flow cytometry
