I use this a lot for immunohistochemistry. I do a lot of neural tracing to map different circuits by using different viral vectors. In order to trace the pathways, I stain for the fluorescent protein expressed by the virus. These projects require DAPI and a mounting medium that produces the best final images. I have used several different types of mounting medium and this one is by far the best. It never leaves bubbles, the DAPI works well, and the hard-set property makes it dry really well.

Protocol Overview

I use a standard immunohistochemistry protocol. After the final wash, I mount the tissue onto slides and allow these to dry for at least one hour. I then coverslip the slides with this mounting medium and I very rarely see bubbles compared to other mounting mediums.

Tips

Briefly dip the well-dried slide in water before applying this mounting medium and coverslipping. The tissue will remain on the slide and the DAPI dilution is not a problem.
