Our lab has transgenic mice that express β-galactosidase on specific cell types. We needed to use this X-gal substrate kit to visualize the β-galactosidase in our fixed mouse retina tissue.

Fixed mouse tissue
After fixing, the mouse retina tissues were incubated with 500uL X-gal solution (100mM in DMSO:Water) in a humidified chamber at 37 degrees over night. The tissue was then fixed again with 4% PFA at room temperature for 30 minutes, then washed with PBS 4 times. The images were capture using a light microscope.
Titrate the amount of X-gal needed for your sample type.
We are able to very clearly visualize the β-galactosidase tagged cells in our mouse tissue.

Cannot combine with fluorescent markers
This is a great assay kit for visualizing β-galactosidase clearly in fixed mouse tissue.
