The overall goal of my research is to gain insights into HIV transmission. The specific goal of this project is to attempt to isolate CD4+ T cells from the tissues following enzymatic and mechanical digestion.This kit was chosen as it is suppose to be good for use in nonhuman primates.
Cell Isolations from the tissue
Rhesus Macaque tissues
Tissues were digested in presence of enzymes, elevated temperature and fine scissors mincing. Miltenyi kit was used according to manufacturer's protocol to isolate CD4 positive T cells from cell suspensions. The success of cell separation was monitored both by fluorescence microscopy by staining for CD4 positive T cells and by flow cytometric analysis by staining for CD45 positive cells.
Filter cells a bit at a time to prevent filter clogging.
Very efficient cell separation was achieved. Greater than 90 percent of cells were CD4 positive CD3 cells.
