After struggling with transfection of HUVECs using standard methods, the Lonza 4D nucleofection system was a godsend. Using the system I am able to to get high transaction efficiency with high viability. This is far superior to liposome based transfections

Protocol Overview

Cells are trypsinised and, washed in PBS, spun down and resuspeneded in the transfection solution. Nucleotide is added (siRNA/DNA)

Tips

Work quickly, the less time the cells spend in transfection buffer the better. Have warm media ready. Cuvettes can be rinsed with H2O and reused.
