Our lab isolated cells from various animal and human tissues for cryo-preservation and downstream applications. We frequently need to count live cells before freezing or other uses (such as RNA/DNA extraction, flow cytometry, and adoptive transfer).

Protocol Overview

10uL of single cell suspension is added to 10uL of trypsin blue. 10uL of this solution is added to a hemocytometer with cover slip. The hemocytometer is placed on the microscope, the light is turned on, and the coarse and fine focus knobs are used to focus the samples before the cells are counted.

Tips

Turn off bulb when finished to preserve it's life.
